I just started playing the game, once you build an immigration center and have reached, lets say 100 people, how do you stop them from coming?

In the Colony Control Panel, click the Policies tab, and check the box for Immigration Ban. No one will come as long as that is checked.

Correct. You can also just recycle your Immigration Centers :)
