Is it allowed to make something like upgrades, lasts from one play to another? 

For example, player get a sword in one play, after 20 seconds it ends and in new play he will still have the sword.

In general we’re looking for games at are ‘complete’ in 20 seconds whereas that mechanic sounds iterative—with the current loop depending on the previous, hence the game isn’t really ‘complete’ in 20 seconds but a longer game requiring several loops to achieve the optimal outcome.

Randomizing the levels, enemy placement, powerup availability etc each play to provide variation keeps the spirit of the jam, but the iterative design not so much.

If you do want to explore that mechanic anyway, your game wouldn’t be removed on those grounds
