There isn't a clear answer here, sorry.

Games are certainly welcome to be submitted in any size, but part of the selection criteria is "How easy will it be to print?" So, while it is an advantage to be A5 already, this isn't absolutely necessary (especially if it's already a similar format such as A4).

If an A4/Letter sized game is selected, would the creator have the opportunity to reformat it to the appropriate size?

That's right. If a game is selected, creators will have an opportunity to resize (and make any final changes) before it goes into print. If you're not able to resize, it will be done by Long Tail by simply scaling. In fact, this is the only modification we are allowed to make to the work.
