This one is patch 0.36 and newer, notice the background was transparent while older version was completely black

1. Try holding the left CTRL. This will skip all dialogs. Let me know if it worked for you. It is worth noting that in the settings all parameters must be enabled as in your screenshot.
2. Also - do not run the game in the launcher, this will lead to a bunch of errors through no fault of ours. The latest version of itch launcher has some kind of bug that breaks not only our game but also other games. Launch the game from the game folder manually.
The problem is :
1. I do not used Itch Launcher, I just play the game directly from your download links
2. You misunderstood about point 1, what I mean is : if you Click and HOLD CTRL at same time, it caused conflict between skip and clicking, thus clicking will be disabled as long as I hold the left CTRL before clicking to anything on the screen. This was not an issue at older patches, but 0.36 and newer have this issue whenever I did that (Holding CTRL while tried to click on the screen)
