For the music, we had an invisible object that played the initial background music, then swapped music routines as the timer started to drain. We would then switch to the tense music when the timer ran out and the ghost started chasing the player. As for the lightning, we simply swapped between two different background color palettes of varying brightness every few seconds using GB Studio's Color mode.
